Expertise and Research Interests

Dr. Bevan's research interests are in two main areas: 1) The development of T cells in the thymus and in particular how the forces of positive selection and negative selection (central tolerance) shape the repertoire of antigen receptors, and 2) The response to foreign antigen by mature T lymphocytes in the periphery.

We believe that most T cells with receptors bearing high affinity receptors for self antigen are efficiently removed by the processes of central tolerance and peripheral tolerance, the latter serving as a backup mechanism. This is true even for tissue restricted self antigens expressed in only one or a few peripheral tissues such as the pancreas or melanocytes. However, tolerance spares low avidity T cells specific for self. Since most tumor target antigens for T cells are non-mutated self antigens, the efficacy of T cell immunotherapy is limited. Our goal is to adapt an in vitro system that can already generate large numbers of immature T cells from blood or marrow stem cells so that differentiation of mature T cells occurs. This generation of mature T cells will happen in the absence of tissue restricted target antigens and we hope to devise means to skew the positively selected T cell repertoire to high affinity reactivity with potential tumor antigen targets.

We have shown that CD4 help is required for the generation of long-lived, functional CD8 memory T cells. One component of what CD4 T cells provide is IL-2. CD8 T cells lacking CD25 (and therefore the high affinity IL-2 receptor), generate memory cells which are unable to mount a robust response to second encounter with antigen. We have found that IL-2 signals can be provided to these cells by the use of potent IL-2/anti-IL-2 mab complexes. Our future goals are to elucidate the role CD4 T cells play in the CD8 T cell response, and to explore the timing of IL-2 signals.
