Expertise and Research Interests

Childhood obesity is predictive of adult obesity and the degree of ponderosity in adults has been shown to be an important predictor of morbidity as well as mortality from atherosclerotic cardiovascular disease. A family study was conducted in Muscatine,Iowa of 1,580 individuals in 284 families to evaluate the genetic contribution to familial clustering of levels of the body mass index (BMI) and found evidence that the segregation of two alleles at a recessive locus explains 35%, while polygenic loci explain 42% of the variability in BMI in this population.

Several rodent models of recessive obesity have also been identified and the traits have been mapped to rodent chromosomal regions which are synthenic with regions on human chromosomes 1 and 7. These results suggest that a search for human genes that are responsible for the variability in BMI is now an important venture.

Our current focus is on answering the question: ''Are there human genes or DNA sequences which have measurable effects on quantitative indices of obesity or on quantitative traits that are associated with obesity among families from Muscatine?'' This study should add to an understanding of the genetic factors involved in the determination of obesity and may lead to early identification of individuals and families at high risk for the chronic diseases that are associated with obesity.

Atherosclerotic heart disease has been shown to have its origins in childhood and young adult life. The Muscatine Study of school-age children, which began almost 25 years ago, provides a unique longitudinal population for study of coronary risk factors obtained during childhood and young adult life with repect to their predictive value for the development and progression of the atheroscleroticprocess in the fourth decade of life.

We are currently examining the relationship of coronary risk factor levels measured in childhood and young adult life to the presence and quantity of coronary artery calcification, a marker of the atherosclerotic process. Results from these investigations may establish whether coronary artery calcification, detected in young adult life, is an indicator in asymptomatic young adults of accelerated atherosclerosis.

In humans, excess blood pressure and body size are associated with increased cardiovascular risk. The association between hypertension and obesity is well established, however, the pathophysiologic relationship remains poorly understood. Both hypertension and obesity are multifactorial phenotypes whose etiologies clearly involve the effects of major genes and polygenes, as well as the effects of numerous environmental factors.

We are using measured-genotype and marker-association methods of analysis to estimate the magnitude of the effects of STRP loci at 20 cM intervals throughout the genome on the determination of blood pressure and body size in 500 unselected Muscatine nuclear families. We are also using affected-pedigree-member methods of linkage analysis to identify genetic loci from throughoutthe genome in a Bedouin tribe with multiple hypertensive members.
