Qualifications

Ph.D., University of Michigan, Biological Chemistry, 1968.

Expertise and Research Interests

My expertise is in the following areas:
Epithelial differentiation (oral and skin) and formation of the protective body surface including
Keratins and Filaggrin.

A second area of expertise is in antimicrobial peptides made by epithelia; regulation of their expression, function, and role in host innate defense in the oral cavity, skin, and mucosa.

Other Expertise

University of Washington, School of Dentistry, Comprehensive Center for Oral Health Research, Scientific Director

Future Research

The role of antimicrobial peptides, including beta-defensins, in oral health is being investigated using cell biology, molecular biology, and genetic approaches, coupled with clinical findings. Future studies will test the hypothesis that individual expression of antimicrobial peptides is correlated with susceptibility to infection. This relationship may be particularly important in populations with immunosuppression due to disease or to drug-related immunosuppression associated with cancer therapy, organ transplant, or autoimmune disorders.

Industrial Relevance

Peptide antibiotics are potentially useful new therapeutic agents that can be used topically. It should also be possible to enhance their synthesis locally by use of regulatory factors. Genetic variation in defensins (single nucleotide polymorphisms or SNPs) can be assayed and used to identify people at risk for infection, as more information becomes available in epidemiological studies on the relationship of defensin SNPs and disease susceptibility. Levels of antimicrobial peptides in saliva may also be a useful measure of risk for susceptibility to infection.
