Expertise and Research Interests

My lab studies the basic adhesive interactions required for cellular growth and movement, with particular emphasis on integrins and their ligands. Under normal conditions, adhesive interactions control tissue development and maintain mature tissue integrity. During wound repair, adhesive interactions change to facilitate healing and remodeling. In diseases such as atherosclerosis, cancer and renal tubulointerstitial fibrosis, cellular growth and movement are aberrant, leading to invasion and pathologicalaccumulation of cells and their byproducts. A similar phenomenon is observed in reponse to surgical tissue implants. We are investigating the role of specific adhesive ligands, in particular secreted products such as
osteopontin and other extracellular matrix proteins, and integrins under these four conditions in vasular and renal models of normal homeostasis, regeneration, and disease. In addition, we are studying adhesive mechanisms required for both normal and tumor angiogenesis. Our studies include those aimed at defining the molecular mechamisms which regulate adhesive protein and integrin expression at the level of the gene, adhesive protein structure/function studies, adhesive protein function in endothelial cells, in vivo function of adhesive protein in vascular and kidney disease, and use of adhesive substrates for biomaterial development.
A final area of the lab investigates molecular mechanisms of ectopic calcification, focussing on both inductive and inhibitory aspects. Such studies are aimed at elucidating active processes controlling medically relevant calcifications such as that occurring in native and bioprosthetic valves.
