Research Interests and Activities: The pathogenesis of several viral infections: Acute viral encephalitis: We are studying the mechanism of viral neurovirulence and the role of the immune response in recovery from viral encephalitis. Sindbis virus belongs to the alphavirus group of mosquito-borne causes of viral encephalitis, and its virulence is largely determined by the viral surface glycoproteins. Recombinant viruses have been constructed to determine which changes in individual amino acids and nucleotides are important for neurovirulence. Antibody is the primary means by which virus is cleared from infected neurons; the mechanism of this clearance is being determined using monoclonal antibodies, immunodeficient mice, and neuronal cultures. Unique aspects of the mononuclear inflammatory response within the nervous system during infection are being examined by flow cytometry and functional analysis of cells.Measles: Measles is associated with immune suppression, as manifested by depressed delayed-type hypersensitivity in vivo and by suppressed proliferation of lymphocytes in vitro. Monocytes are infected with measles virus and the effects of this infection on immune function are being studied. Proliferative and cytokine responses of lymphocytes to live and inactivated measles virus are being determined.HIV: The pathogenesis of nervous system disease in AIDS is being studied through prospective studies of the clinical features of HIV-infected patients, of cerebrospinal fluid and leukocytes, and of tissues taken at autopsy.
