Expertise and Research InterestsMy current research interests and areas of expertise include taxonomy and the systematics of thermophilic and extreme thermophilic anaerobic bacteria as well as their physiological and biochemical properties especially in respect to biotechnological application. Presently we are investigating especially novel alkalithermophilic anaerobes including extreme halophilic alkalithermophiles, which represent the novel Order Natranaerobiales in the Phylum Firmicutes. An area of interest is the biogeography and biogeochemistry of thermophiles, i.e., how microorganisms influence the geochemistry and mineralogy of geothermal environments and vice versa. We investigate thermostable enzymes from anaerobic thermophiles which included mainly in the past hemicellulases, cellulases,and proteolytic enzymes. Presently we are working on the most thermostable and thermophilic lipases from Thermosyntropha lipolytica as well as in cooperation with Prof Shao (China) on the regulation of the ethanol-DH in Thermoanaerobacter. We are interested in temperature effects on physiological properties including but not limited to growth rate, growth range (we developed a specific theory on bacteria growing over a wide temperature span), enzyme stability, spore heat stability, and bioenergetic of the halophilic alkalithermophiles
Presently, we are not anymore working actively on the anaerobic degradation of halogenated aromatic compounds (such as poly chlorinated biphenyl (PCB's and 2,4-dichlorophenols) or on the enzymes catalyzing the reversible co-factor independent anaerobic decarboxylation/carboxylation of aromatic compounds
Two sideline: 1)(although presently no ongoing research project by a graduate student or postdoc) is the old research area of N2-fixation by facultative chemolithoautotrophic and microaerophilic N2-fixing bacteria. This includes the taxonomic systematic, physiological and biochemical properties as well as plant microbe interactions of Xanthobacter with patty rice. 2) anaerobic mesophilic microorganisms from acid mine drainage passed through a constructed wetland model system Other ExpertiseHorticulture: 4 years of experience in Germany prior to study of chemistry and microbiology. Future ResearchOngoing research deals with: - The diversity of anaerobic thermophiles, with the focus on alkalithermophiles and halophilic alkalithermophiles - Anaerobic hemicellulose degradation at elevated temperatures - The elucidation of the bioenergetics of the haloalkalithermophiles -developing a genetic system for Natranaerobius using our previously developed sytem for Thermoanaerobacterium - biogeography of Thermoanaerobacter uzonii in the Uzon Caldera (Kamchatka, Far East Russia) Industrial RelevanceMany of our novel thermophilic isolates have quite a lot of potential for industrial applications either as source for genes for artificial evolution of novel enzymes or directly as biocatalyst. (proteases, lipases, hydrolytic enzymes for biomass degradation). The highly thermophilic lipase has a potential in the position modification of lipids and synthesis of specific lipids and synthesis of higher fatty acid esters. Our research on the influence of environmental factors on the anaerobic (reductive) dehalogenation of PCBs and chlorophenols are important for development of bioremediation processes and/or for estimating the recalcitrance of the compounds in a given environment. Our data can be used to help to decide whether bioremediation should be done or whether the environment will slowly remove the compounds (passive bioremediation).
