Expertise and Research InterestsMy basic research interests are in molecular aspects of host - parasite interactions, focusing on Mycoplasma pneumoniae. M. pneumoniae is a pathogen of the human respiratory tract, causing bronchitis and atypical or 'walking' pneumonia. This cell wall-less prokaryote is the leading cause of pneumonia in older children and young adults. M. pneumoniae colonizes the mucosal epithelium of the bronchial passages via a differentiated terminal structure called the attachment organelle, which also functions in gliding motility and cell division. We study the architecture, assembly, regulation, and function of this terminal organelle. Ultrastructural and molecular approaches are used to dissect the adherence apparatus in order to define more clearly the identities and roles of its component proteins in attachment organelle function. By characterizing a battery of mutants defective in attachment organelle assembly we have begun to reconstruct the assembly process. Transposon-based approaches are being utilized to manipulate the mycoplasma genome in order to probe this process further, including structure-function analyses of attachment organelle proteins. In addition, fluorescent protein fusions are being employed to examine the assembly process in real time. Finally, gliding motility yields satellite colonies when wild-type M. pneumoniae is grown under the appropriate culture conditions. Non-motile mutants fail to produce satellite growth under such conditions. We are utilizing a combination of approaches to dissect the mechanism of gliding motility and assess the contribution of gliding to M. pneumoniae virulence.
