Expertise and Research InterestsOver 34 million Americans have decreased bone mass and an additional 10 million are classified as osteoporotic. Aging and disuse (spaceflight, spinal cord injury, limb fixation) can result in bone loss and its associated increase in fracture risk, which is strongly associated with functional dependence, depression, and increased morbidity with the elderly. Osteoblasts (the cells that make bone) and osteocytes (mature osteoblasts embedded in mineralized bone) are involved in responses to skeletal loading and coordination of bone resorption and formation. The function of these cells is in part controlled by the expression of runx2, a transcription factor that is important for the expression of genes associated with mature osteoblast and osteocyte phenotype and function. Using cellular and molecular approaches we are identifying mechanisms accounting for decreased bone mineral density associated with aging, spaceflight and diseases such as type I diabetes. Specifically, we find that these conditions modulate MAPK signaling pathways, transcription factor activities (AP-1, CRE, runx2), stress proteins and gap junctions, leading to the suppression of osteoblast differentiation. Ongoing studies will further identify specific mechanisms regulating bone formation in vitro and in vivo under stress and disuse conditions and contribute to the development of treatments to enhance bone formation under conditions of fracture healing, bone implants and osteoporosis. Other ExpertiseMy lab is using a combination of molecular biology, cell biology, and physiology to address mechanisms regulating bone cell growth and differentiation. These approaches and experimental techniques include: protein, RNA and DNA isolation and analyses; gene and cell cloning; stable and transient transfections; proteomic and genomic analyses; quantitative real time RT-PCR; immunofluroescence; electrophoretic mobility shift assays; culturing of primary bone cells, marrow cells, cell lines, and tumor cells; mouse models of bone loss; histology; microcomputed tomography for 3D visualization and measurement of bone parameters; human bone analyses. We are also working on several interdisciplinary projects with chemistry, biochemistry, engineering, animal science, and orthopedics to enhance bone formation in bone implants.

Industrial RelevanceIdentification of novel mechanisms regulating bone formation can contribute to the development of new drugs targeted at specific signaling pathways to enhance bone formation under conditions of bone repair (fracture, bone implants) or bone loss associated with disease or aging.
