Expertise and Research Interests

Research interests:

Platelet kinetics and function; physiology of platelets in disease: Bench and clinical research studies of platelet turnover using unique platelet markers developed in this laboratory. These investigations study changes in plateletactivation, tyrosineprotein kinases, and platelet RNA content. Functional aspects of
platelet studies include phosphatidylserine expression, leukocyte-
platelet conjugate formation, and platelet adhesion under shear
conditions.

Mechanisms of hemostatic and inflammatory changes with extracorporeal circula
tion and in the physiology of cardiopulmonary bypass. In vitro, ex vivo, and in vivo studies of cellular and soluble changes in peripheral blood during artificial biomembrane contact with blood.This involves the study of a) biomaterial compatibility, b) novel anti-coagulant and anti-inflammatory compounds to block blood cell and complement activation, and c) development of new models of shear stress effects on flowing whole blood, including anti-Xa inhibitors,
complement blockade, novel anticoagulants, and thrombin inhibitors.
Signal transduction and secondary effects of platelet-leukocyte-endothelial adhesion: Studies of ligand receptor interactions between heterotypic blood cells are conducted in in vivo an
d in vitro settings to determine functional properties related to secondary signaling, cytokine elaboration, and subsequent changes in receptor affinity and cellular function. These studies are related to platelet transfusion in vivo, stem cell transplantation in a murine model, immune thrombocytopenia, lupus, antiphospholipid syndrome, and hypercoagulable or prothrombotic states.

Studies of platelet and complement activation relevant to models of platelet clearance, thrombocytopenia, and hypercoagulable states.

Industrial Relevance

We have particular expertise in blood cell analysis and novel methods for determining cell turnover and production by non-invasive peripheral blood assays. These are relevant to automated cell counting, laboratory hematology instrumentation, and rapid point-of-care diagnostic testing. Our expertise also extends to novel coagulation assays, including cell-free and cell-dependent systems, shear stress models, and extracorporeal circuits.
