Expertise and Research InterestsPneumonectomy (removal of one lung) results in rapid compensatory growth of the remaining lung. The mechanisms that regulate this growth are unknown, and the long-term goal of my research is to understand the molecular mediators that trigger and regulate compensatory lung growth. Recently we have evaluated the impact of exogenous administration of retinoic acid, epidermal growth factor and keratinocyte growth factor on lung growth. We are currently studying the role of nitric oxide on compensatory lung growth. We have demonstrated that compensatory lung growth is impaired in eNOS- and iNOS-knockout mice, and we have shown that epidermal growth factor receptor and vascular endothelial growth factor (VEGF) are upregulated during compensatory lung growth. Our studies are directed at testing our overall hypothesis that nitric oxide is a key mediator of compensatory lung growth through the modulation of inflammation, angiogenesis and type II alveolar cell proliferation. Gene array studies are being conducted to help identify genes which play critical roles in compensatory lung growth. We are also conducting studies evaluating unequal growth among the lobes of the lung by measuring vascular growth, epithelial and endothelial proliferation and blood flow.
A second area of research in my lab is the study of lung ischemia-reperfusion (IR) injury. We have demonstrated that lung IR injury occurs as a biphasic response in which the acute phase is macrophage-dependent followed by a neutrophil-dependent phase of injury. This finding is important because it reinforces our concept that resident lung cells (i.e. cells residing in the donor lung rather than circulating leukocytes from the recipient) initiate IR injury upon transplantation. We have also demonstrated the rapid nuclear activation of NF-kappaB following lung transplantation and have shown that inhibition of NF-kappaB activation (during lung preservation) improves lung function and diminishes inflammation. We are now utilizing both a mouse in-situ, buffer-perfused, isolated lung model as well as an in vivo hilar clamp model of lung IR injury. Using various mouse knockout strains with these two models will we aim to determine how lung epithelial cells respond to IR, how they interact with alveolar macrophages, and how they participate in recruitment of neutrophils during IR injury.
In vitro studies are also being conducted using mouse macrophage cells (RAW) and type 2 alveolar epithelial cells (MLE-12) to evaluate the response of these cells to hypoxia-reoxygenation.
